Stimulated by Tamoxifen and Inhibited by the Pure Insulin-like Growth Factor I Gene Expression in the Uterus Is

چکیده
Estrogen-induced uterine insulin-like growth factor I (IGF-I) expression has been demonstrated to mediate at least in part the uterotrophic action of estradiol. We studied the effects of tamoxifen, a partial antagonist to the estrogen receptor widely used in the treatment of breast cancer, and ICI 182780, a pure antagonist to the estrogen receptor, on uterine weight and uterine IGF-I gene expression in the rat. Tamoxifen increased uterine weight to 125% of control values and doubled uterine IGF-I expression. In contrast, ICI 182780 reduced uterine weight to 60% of control and uterine IGF-I gene expression to 13 % of control. These results demonstrate for the first time that uterine IGF-I expression is a molecular marker that correlates with the effects of partial agonists and antagonists to the estrogen receptor on the uterus. Furthermore, the induction of uterine IGF-I expression by tamoxifen provides a molecular mechanism to account for the uterotrophic effects which are commonly seen with tamoxifen therapy and which have been associated with endometriai neoplasia.

متن کاملEffects of Inflammatory Cytokine Tumor Necrosis Factor-α on Human Mesenchymal Stem Cell Gene Expression: A Mechanism for Liver Regeneration
Introduction Insulin-like growth factor I (IGF-I) which is produced in abundance in the normal adult liver, is deeply involved in hepatocyte survival, growth, and differentiation during liver development. IGF-I plays the roles via the receptor (IGF-IR) signaling pathway. IGF-IR unlike IGF-I is expressed strongly in the developing liver, but much more weakly in adults. Objective: We hypothesi...
متن کاملP-64: The Relationship between Polymorphism in Gene of Insulin-Like Growth Factor-I and The Serum Periparturient Concentration in Holstein Dairy Cows
Background: One of the most important metabolic factors affecting the reproductive activity is insulin-like growth factor-I (IGFI) concentration changes after calving. Recently, Maj et al. (2008) discovered a significant association between the IGF-I genotypes based on the 5'-untranslatedregion (5'-UTR) of IGF-I gene and the IGF-I blood level. The objective of this study is to investi...
متن کاملEffects of zinc sulfate (ZnSO4) in diet on the growth performance and gene expression of Insulin-like growth factor-1 (IGF-1) and Ghrelin in goldfish (Carassius auratus)
The zinc element (Zn) is one of the micronutrients and an essential element of the body which has some deleterious and irreparable effects on growth in case of lack it. Thus, we aimed to assess IGF-1 in goldfish with Zn deficiency and to investigate the effect of Zn supplementation on these parameters. Goldfish (Carassius auratus) juvenile (3.5 g) were fed purified diets based on casein as ...
متن کاملANTISENSE RNA TO THE TYPE I INSULIN-LIKE GROWTH FACTOR RECEPTOR REVERSED THE TRANSFORMED PHENOTYPE OF PC-3 HUMAN PROSTATE CANCER CELL LINE IN VITRO
The insulin-like growth factor I receptor (IGF-IR) plays an essential role in the establishment and maintenance of transformed phenotype. Interference with the IGF-IR pathway by antisense causes reversal of the transformed phenotype in many rodent and human tumor cell lines. We stably transfected the PC-3 human prostate cancer cell line with an IGF-IR antisense RNA expression plasmid. The ...
